研究概述
RNA转录和DNA复制是生命过程中必须发生的两个核心生物学事件。然而,我们前期的研究工作意外地揭示,无时无刻进行着的RNA转录可以通过形成R-loop干扰DNA复制叉运行, 进而影响基因组稳定性。维持基因组稳定是生命得以存在和延续的根本。毫无疑问,机体必然相应地发展形成一套有效的应对机制以维护基因组稳定。本实验室的主要研究兴趣在于运用遗传学,分子生物学,生物化学和生物信息学等方法,理解哺乳动物细胞是如何应对在生长和增殖过程中似乎是必然会出现的转录、复制和基因组稳定性,以维护三者间的冲突;并探索这一冲突形成后可能产生的病理后果。希望我们的研究能有助于加深对于肿瘤等重大疾病发病机理的理解,从而为癌症的诊断、治疗和药物的开发提供新的理论基础。
目前实验室正在开展的工作包括:
1. R-loop破坏基因组不稳定性的应对机制
2. 转录在肿瘤发生以及肿瘤细胞染色体不稳定性建立过程中的作用
3. 染色体脆性表达的分子机理
4. 哺乳动物细胞中停顿复制叉的稳定和重新启动的分子机制
5. 人基因组DNA复制起始的时、空调控机制
6. RNA加工过程在癌症发生和发展中的作用
